PAX3 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry (Paraffin) analysis of PAX3 was performed in paraffin-embedded human esophageal carcinoma tissue using PAX3 Monoclonal Antibody (GT1210) (Product # MA5-31583) at a dilution of 1:200. Antigen Retrieval: Citrate buffer, pH 6.0, 15 min.

PAX3 Antibody in Western Blot (WB) — Western blot was performed using Anti-PAX3 Monoclonal Antibody, (Product # MA5-31583) and a ~52 kDa band corresponding to PAX3 was observed across the panel tested except T-47D and MCF-7 which are reported to be negative. An uncharacterized band (*) at ~110kDa was also observed across the panel tested. Modified Whole cell extracts (1% SDS) (30 µg lysate) of A-375 (Lane 1), U-87 MG (Lane 2), T-47D (Lane 3), MCF-7 (Lane 4), Neuro-2a (Lane 5), PC-12 (Lane 6) and NIH/3T3 (Lane 7) were electrophoresed using Novex® NuPAGE® 4-12% % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005)..

PAX3 Antibody — Antibody specificity was demonstrated by CRISPR-Cas9 mediated knockout of target protein. A loss of signal was observed for target protein in PAX3 (KO) cell line compared to control cell line using Anti-PAX3 Monoclonal Antibody (Product # MA5-31583). An uncharacteristic non-specific band at ~110 kDa was observed.

Target Information

Pax 3 (Paired Box 3) is a transcription factor that is involved in development of the peripheral nervous system, melanocytes, some vascular smooth muscle, and a number of other derivatives. It regulates neurogenesis in pre-migratory neural crest cells from the dorsal neural tube, and in myogenic progenitors in the presomitic mesoderm and the hypaxial somites. Alternative splicing results in isoforms having different C-termini.
