Immunofluorescent analysis of PC2 in U2OS cells using a PC2 polyclonal antibody (Product # PA1-31853). The image reveals the expected discrete nuclear structure that is termed the PcG body corresponding to the known localization of PC2. Cells were fixed in PBS with 4% PF for 5 min, permeabilized with 0.5% Polyethylene glycol tert-octylphenyl ether-PBS for 5 min, and blocked with 5% milk/0.2% Tween for 1 hr. Primary antibody was used at 1:200 in 5% milk/0.2% Tween for 1 hr and secondary antibody was incubated for 30 min. All blocking and incubation steps were carried out at 37°C. Nuclei were counterstained with Hoechst stain (blue).
