Western blot analysis was performed on nuclear enriched extracts (30 µg lysate) of HeLa (Lane 1), MOLT-4 (Lane 2) and SH-SY5Y (Lane 3). The blot was probed with Anti-PIAS1 Mouse Monoclonal Antibody (Product # 39-6600, 2 µg/ml) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.4 µg/ml, 1:2500 dilution). A 72 kDa band corresponding to PIAS1 was observed across the cell lines tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 10 % Bis-Tris gel (Product # NP0302BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

PIAS1 functions as an E3-type small ubiquitin-like modifier (SUMO) ligase, stabilizing the interaction between UBE2I and the substrate, and as a SUMO-tethering factor. This protein plays a crucial role in transcriptional coregulation of various cellular pathways, including the STAT pathway, the p53 pathway and the steroid hormone signaling pathway. It functions in testis as a nuclear receptor transcriptional coregulator and may have a role in androgen receptor initiation and maintenance of spermatogenesis. The effects of transcriptional coregulation, transactivation or silencing, may vary depending upon the biological context.
