Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is Hodgkin's lymphoma.
Phosphoinositide-specific phospholipase C (PLC) plays a critical role in the initiation of receptor mediated signal transduction through the generation of the two second messengers, inositol 1,4,5-triphosphate and diacylglycerol from phosphatidylinositol 4,5 bisphosphate. There are many mammalian PLC isozymes, including PLC beta1, PLC beta2, PLC beta3, PLCbeta4, PLC?1, PLC?2, PLC d1, PLCd2 and PLCe with molecular weights ranging from 85 to 150kDa.
