PNK Antibody (MA5-44764) in ICC/IF
Immunocytochemistry analysis of PNKP in HeLa cells. Cells were fixed in 4% paraformaldehyde for 10 minutes at 37 , permeabilized with 0.05% Triton X-100 in PBS for 20 minutes, and then blocked with 2% negative goat serum for 30 minutes at room temperature. Samples were incubated in PNKP Monoclonal antibody (Product # MA5-44764) using a dilution of 1:100 in 2% negative goat serum overnight at 4 followed by Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) secondary antibody at a dilution of 1:1,000. Nuclear DNA was labelled in blue with DAPI.

Target Information
Anti-human Polynucleotide Kinase Mouse Monoclonal Antibody recognizes polynucleotide kinase (PNK), a 57 kDa nuclear enzyme implicated in DNA strand repair processes that occur during DNA replication and recombination, or in response to DNA damaging agents. PNK targets the DNA 5'-hydroxy and 3'-phosphate termini for phosphorylation and dephosphorylation events. During strand breakage in human cells, normal PNK associates with other nuclear proteins to prepare the DNA termini for the enzymatic actions of DNA polymerase-beta and DNA ligase III. PNK is a key functional component in maintaining the genetic stability of human cells.
