PTPN11 Antibody in Western Blot (WB) — Equivalent amounts of cell lysates (10 µg per lane) of wild-type 293T cells (WT, Cat# LC810293T) and PTPN11-Knockout 293T cells (KO, Cat# LC811214) were separated by SDS-PAGE and immunoblotted with anti-PTPN11 monoclonal antibody TA501914, (1:500). Then the blotted membrane was stripped and reprobed with anti-beta-actin antibody (TA811000) as a loading control.

PTPN11 Antibody in Flow Cytometry (Flow) — Flow cytometric analysis of Jurkat cells, using anti-PTPN11 antibody (TA501914), (Red), compared to a nonspecific negative control antibody (TA50011), (Blue).

PTPN11 Antibody — Antibody specificity was demonstrated by CRISPR-Cas9 mediated knockout of target protein. Loss of signal was observed for target protein in PTPN11 knockout (KO) 293T cell line using Anti-PTPN11 monoclonal antibody (Product # CF501914).

For reconstitution, we recommend adding 100 µL distilled water to a final antibody concentration of about 1 mg/mL. To use this carrier-free antibody for conjugation experiments, we strongly recommend performing another round of desalting. (Zeba Spin Desalting Columns, 7KMWCO, 0.5 mL, Product # 89882)

Target Information

The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains two tandem Src homology-2 domains, which function as phospho-tyrosine binding domains and mediate the interaction of this PTP with its substrates. This PTP is widely expressed in most tissues and plays a regulatory role in various cell signaling events that are important for a diversity of cell functions, such as mitogenic activation, metabolic control, transcription regulation, and cell migration. Mutations in this gene are a cause of Noonan syndrome as well as acute myeloid leukemia.
