Immunofluorescent analysis of Phospho-EIF2A pSer52 using a Phospho-EIF2A pSer52 polyclonal antibody (Product # PA1-26686). Left: rat brain (mediodorsal neocortex) sections following traumatic axonal injury (TAI) (24 h). Control (prior to injury induction) shows low basal levels of EIF2 alpha. Right: Image shows double labeling of APP (red) and EIF2 alpha (green) in neurons subjected to traumatic axonal injury (TAI). Inset (above): higher magnification of neuronal soma with TAI. EIF2 alpha immunofluorescence is localized in cytoplasm of neuronal cell body.

PA1-26686 detects eIF2A (phospho S52) from human and mouse samples. PA1-26686 is expected to cross react with a wide range of other species due to sequence homology.
PA1-26686 has been successfully used in Western blot procedures.
The PA1-26686 immunogen is a synthetic phosphopeptide corresponding to a portion of human eIF-2 alpha containing serine 52. This portion is conserved among many species including rat, pig, cow, fruit fly, and yeast.
The translation initiation factor EIF2 catalyzes the first regulated step of protein synthesis initiation, promoting the binding of the initiator tRNA to 40S ribosomal subunits. Binding occurs as a ternary complex of methionyl-tRNA, EIF2, and GTP. EIF2 is composed of 3 nonidentical subunits, the 36-kD EIF2-alpha subunit (EIF2S1), the 38-kD EIF2-beta subunit (EIF2S2; MIM 603908), and the 52-kD EIF2-gamma subunit (EIF2S3; MIM 300161). The rate of formation of the ternary complex is modulated by the phosphorylation state of EIF2-alpha (Ernst et al., 1987 [PubMed 2948954]).
