To expose target proteins, antigen retrieval was performed using 10 mM sodium citrate (pH 6.0), microwaved for 8-15 min. Following antigen retrieval, tissues were blocked in 3% H2O2-methanol for 15 min at room temperature, washed with ddH2O and PBS, and then probed with a Phospho-FAK2 /PYK2 pTyr402 (17HCLC) Oligoclonal antibody (710087) diluted in 3% BSA-PBS at a dilution of 1:20 overnight at 4°C in a humidified chamber. Tissues were washed extensively in PBST and detection was performed using a HRP-conjugated secondary antibody followed by colorimetric detection using a DAB kit. Tissues were counterstained with hematoxylin and dehydrated with ethanol and xylene to prep for mounting.

PyK2, also known as protein tyrosine kinase, is known to be expressed in the central nervous system and from cells derived from hematopoietic lineages. In response to various stimuli that increase the intracellular calcium concentration, PyK2 gets rapidly phosphorylated on the tyrosine residues. On activation of the Pyk2 protein, the MAP kinase signaling pathway is activated and its activation brings about modulation of the ion channel function. The phosphorylation of tyrosin is brought about by reactions involving protein tyrosine phosphatases and the RAFT/Pyk2 kinase proteins.
