Immunofluorescence analysis of FoxA2 [pT156] was done on 70% confluent log phase HepG2 cells. The cells were fixed with 4% paraformaldehyde for 15 minutes; permeabilized with 0.25% Triton™ X-100 for 10 minutes followed by blocking with 5% BSA for 1 hour at room temperature. The cells were incubated with ABfinity™ FoxA2 [pT156] Recombinant Rabbit Oligoclonal Antibody (710680) at a dilution of 1:400 in 1% BSA and incubated for 3 hours at room temperature and then labeled with Alexa Flour® 488 Goat Anti-Rabbit IgG Secondary Antibody (A11008) at a dilution of 1:400 for 30 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (S36938). F-actin (Panel c: red) was stained with Alexa Fluor® 594 Phalloidin (A12381). Panel d is a merged image showing nuclear localization of FoxA2 [pT156]. Panel e is no primary antibody control. Panel e shows competition with FoxA2 [pT156] peptide. The images were captured at 20X magnification.

FoxA2 is a transcription factor, which belongs to forkhead family of DNA binding proteins and also a member FoxA subfamily. Apart from the centrally located forkhead box domain, FoxA2 protein has conserved activation domain II and III at the C-terminus and conserved activation domain IV and V are located at the N-terminus. FoxA2 proteins interact with histones H3 and H4 through C-terminal region and displace nucleosomal histones to remodel chromatin and recruit cell-type restricted factors to initiate gene expression that confers cell and tissue type identities. FoxA2 play a key role in regulation of glucose and lipid metabolism and energy balance at the organism level. FoxA2 exert its glucose and lipid homeostasis function by regulating genes at the metabolically active sites such as, liver, pancreatic alpha and beta cells and adipocytes.
