For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM NaF should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background. PA1-84839 detects a band of ~100kDa in rat brain lysates.
GluR1, widely expressed throughout the nervous system, is a component of AMPA receptors (alpha-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid receptors), which play a key role at excitatory synapses. Phosphorylation of Ser831 and Ser845 is reported to potentiate GluR1 function, suggesting a possible role in synaptic plasticity, learning and memory.
