This antibody is reactive to human and rat JNK1 and 2. Other species of JNK1 and 2 have not been tested, and JNK3 (found primarily in neuronal cell lines) has not been detected. The antibody has been negatively preadsorbed using a non-phosphopeptide corresponding to the site of phosphorylation to remove antibody that is reactive with non-phosphorylated JNK1 and 2. The final product is generated by affinity chromatography using a JNK1 and 2-derived peptide that is phosphorylated at threonine 183 and tyrosine 185. Positive controls used: HEK 293 +/- UV irradiation treatment; PC12 cells +/- sorbitol.
C-Jun N-terminal Kinase (JNK) is also known as Mitogen-activated protein kinase (MAPK8). It belongs to the MAPK superfamily of stress-activated protein kinases/MAP kinases (SAPK/MAPK). MAPKs are serine-threonine protein kinases that are activated in response to a variety of extracellular stimuli, and mediate signal transduction from the cell surface to the nucleus. JNK is involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation, and development. JNK pathways are activated by stress and inflammatory signals. JNK is expressed as ten different isoforms due to differential mRNA splicing. The predominant forms are JNK1 (~49 kDa) and JNK2 (~55 kDa). JNK is dually phosphorylated and therefore fully activated by MEK4 and MEK7 on threonine 183 and tyrosine 185 within the activation loop. The JNK and p38 pathways function to modulate cell cycle, apoptotic, and transcriptional responses to stress.
