Phospho-RSK1 (Thr359, Ser363) Antibody in Immunofluorescence (IF) — Immunofluorescence analysis of Phospho-RSK1 (Thr359, Ser363) was performed using 70% confluent log phase HeLa cells treated with TNF-alpha (25ng/ml, 20 minutes) post overnight serum starvation. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with Phospho-RSK1 (Thr359, Ser363) Polyclonal Antibody (Product # PA5-38309) at 1:200 dilution in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing induction of expression upon treatment. Panel e represents control cells without treatment. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

Phospho-RSK1 (Thr359, Ser363) Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of Phospho-p90RSK/RSK1 pThr359/pSer363 in paraffin-embedded human breast carcinoma using a Phospho-p90RSK/RSK1 pThr359/pSer363 polyclonal antibody (Product # PA5-38309).

Phospho-RSK1 (Thr359, Ser363) Antibody in Western Blot (WB) — Western blot analysis was performed on modified whole cell extract (1% SDS) (30 µg lysate) of A431 (Lane 1), A431 treated with EGF (200ng, 15 mins) upon overnight serum starvation (Lane 2), HeLa (Lane 3) and HeLa treated with TPA (200nM, 20 mins) (Lane 4). The blot was probed with Anti-PhosphoRSK1 Polyclonal Antibody (Product # PA5-38309, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). A 81 and 72 kDa band corresponding to both isoforms of RSK1 was observed in cell lines tested and was enhanced upon treatment.

Phospho-RSK1 (Thr359, Ser363) Antibody in Cell treatment — Altered expression of target protein upon cell treatment demonstrates antibody specificity. Western blot analysis of Phospho-RSK1 (Thr359, Ser363) using Phospho-RSK1 (Thr359, Ser363) Polyclonal Antibody (Product # PA5-38309) shows an induction in PhosphoRSK1 upon treatment with EGF and TPA. {TM}

Target Information

This gene encodes a member of the RSK (ribosomal S6 kinase) family of serine/threonine kinases. This kinase contains 2 nonidentical kinase catalytic domains and phosphorylates various substrates, including members of the mitogen-activated kinase (MAPK) signalling pathway. The activity of this protein has been implicated in controlling cell growth and differentiation. Alternate transcriptional splice variants, encoding different isoforms, have been characterized.
