Peptide Competition. Lysates prepared from A431 cells left unstimulated (1) or stimulated with 100 ng/mL PMA (a phorbol ester activator of protein kinase C) for 30 minutes at 37oC (2-5) were resolved by SDS-PAGE on a 10% polyacrylamide gel and transferred to PVDF. Membranes were blocked with a 5% BSA-TBST buffer for one hour at room temperature and incubated with RSK1 [pS380]/2 [pS386] antibody for one hour at room temperature in a 3% BSA-TBST buffer, following prior incubation with: no peptide (1, 2), the non-phosphopeptide corresponding to the immunogen (3), a generic phosphoserine-containing peptide (4), or, the phosphopeptide immunogen (5). After washing, membranes were incubated with goat F(ab"e;)2 anti-rabbit IgG HRP conjugate (Cat. no. ALI4404) and bands were detected using the Pierce SuperSignal™ method. The data show induction of the RSK1 [pS380]/2 [pS386] antibody signal by PMA treatment and that the peptide corresponding to this site blocks the antibody signal, thereby demonstrating the specificity of the antibody.

This gene encodes a member of the RSK (ribosomal S6 kinase) family of serine/threonine kinases. This kinase contains 2 nonidentical kinase catalytic domains and phosphorylates various substrates, including members of the mitogen-activated kinase (MAPK) signalling pathway. The activity of this protein has been implicated in controlling cell growth and differentiation. Alternate transcriptional splice variants, encoding different isoforms, have been characterized.
