OPA1-03091 detects p-Src from human, mouse, and chicken tissues and cells.
OPA1-03091 has been successfully used in Western blot procedures. By Western blot, this antibody detects an ~60 kDa protein representing p-Src from platelet-derived growth factor (PDGF) treated NIH-3T3 cell extract. Previous lots of this antibody have been used in immunostaining.
OPA1-03091 immunizing phosphopeptide was derived from human Src that contains phosphorylated tyrosine 418.
The Src family of protein tyrosine kinases are non-receptor kinases whose role is important in the regulation of growth and differentiation of eukaryotic cells (1,2). Activity of Src is regulated phosphorylation of two tyrosine residues (Tyr 416 and Tyr527). Phosphorylation of these two sites has opposing effects. Phosphorylation of Tyr416 in the activation loop of the kinase domain upregulates enzyme activity. It has been shown that the phosphorylation of Tyr527 decreases enzyme activity.
