S100A9 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of S100A9 was performed using 70% confluent log phase THP-1 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with S100A9 Recombinant Rabbit Monoclonal Antibody (209)(Product # MA5-29545) at 1:100 dilution in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415). Panel d represents the merged image showing cytoplasmic localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

S100A9 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical staining of human S100A9 in human spleen with S100A9 Recombinant Rabbit Monoclonal Antibody (209) (Product # MA5-29545, 1:200, formalin-fixed paraffin embedded sections).

S100A9 Antibody in Western Blot (WB) — Western blot was performed using anti-S100A9 Monoclonal Antibody (Product # MA5-29545) on whole cell extracts (30 µg lysate) of THP-1 (Lane1), Jurkat (Lane 2) and Raji (Lane 3) and 13.2 kDa band corresponding to S100A9 protein was observed inly in THP-1 and absent in Jurkat and Raji. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 0.25 µg/mL, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

S100A9 Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across tissue panel owing to their inherent genetic constitution. Relative expression of S100A9 protein was observed in ThP-1 cell line in comparison to Jurkat and Raji using S100A9 Recombinant Rabbit Monoclonal Antibody (209) (Product # MA5-29545) in western blot. {RE}

Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire.

This antibody has specificity for Human S100A9.

Target Information

S100A9 (MRP-14) is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21. This protein may function in the inhibition of casein kinase and altered expression of this protein is associated with the disease cystic fibrosis.
