Western blot analysis was performed on membrane extracts (30 µglysate) of U-87 MG (Lane 1), Neuro-2a (Lane 2), SH-SY5Y (Lane 3), SK-N-AS (Lane 4), T98G (Lane5), IMR-32 (Lane 6), and tissue extract (30 µglysate) of rat brain (Lane 7). The blots were probed with Rabbit Anti-SCAMP1 Polyclonal Antibody (Product # PA1-739, 2 µg/ml) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). A 36 kDa band corresponding to SCAMP1 was observed across the cell lines and tissues tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

PA1-739 detects secretory carrier membrane protein 1 (SCAMP 1) from human, mouse and rat samples.
PA1-739 has been successfully used in Western blot and immunoprecipitation procedures. By Western blot, this antibody detects an ~31 kDa protein representing SCAMP 1 in rat and mouse brain tissues as well as HeLa cell lysates.
PA1-739 immunizing peptide corresponds to amino acid residues 1-17 from human SCAMP 1. This sequence is completely conserved between human, rat, and porcine SCAMP 1. PA1-739 immunizing peptide (Cat. # PEP-105) is available for use in neutralization and control experiments.
Secretory carrier membrane proteins (SCAMPs) are proteins that are components of post-Golgi membranes. These ubiquitously expressed proteins are implicated to function in membrane trafficking. In fibroblasts, SCAMPs are concentrated in compartments that are involved in the recycling of cell surface receptors and endocytosis. In neurons, SCAMPs are associated with synaptic vesicles, secretion granules and transporter vesicles. Of the three known SCAMPs, SCAMP 1 is the most abundant. SCAMP 1 has been shown to be required for the full execution of stable exocytosis in mast cells.
