SHP-1 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of SHP-1 was performed using 70% confluent log phase Jurkat cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with SHP-1 Mouse Monoclonal Antibody (Product # MA5-11669) at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A28175) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing nuclear and cytoplasmic localization. Panel e shows the no primary antibody control. The images were captured at 60X magnification.

SHP-1 Antibody in Western Blot (WB) — Western blot was performed using Anti-SHP-1 Monoclonal Antibody (1SH01 (11D7C8H5))(Product # MA5-11669) and a 63kDa band corresponding to SHP-1 was observed across the cell lines and tissues tested. Whole cell extracts (30 µg lysate) of Jurkat (Lane 1), THP-1 (Lane 2), HEL 92.1.7 (Lane 3), MCF7 (Lane 4), K-562 (Lane 5), U-2 OS (Lane 6), Mouse Spleen (Lane 7), Rat Spleen (Lane 8), Mouse Skeletal Muscle (Lane 9) and Rat Skeletal Muscle (Lane 10), were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # LC2001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1 µg/mL) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).The bands observed at 25 kDa are light chain IgG bands in the mouse tissue lysates.

SHP-1 Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell lines and tissue tested owing to their inherent genetic constitution. Relative expression of SHP-1 was observed in Jurkat, THP-1, HEL.92.1.7, MCF-7 and Mouse Spleen and Rat spleen, which are high expressing models and was found to be negative in K-562, U-2 OS, Mouse Skeletal muscle and Rat Skeletal muscle, which are low expressing models as reported. using Anti-SHP-1 Monoclonal Antibody (1SH01 (11D7C8H5)) (Product # MA5-11669) in Western Blot. {RE}

Target Information

SHP-1 (PTPN6) is a non-receptor protein tyrosine phosphatase that is expressed primarily in hematopoietic cells. The enzyme is composed of two SH2 domains, a tyrosine phosphatase catalytic domain, and a carboxy-terminal regulatory domain. SHP-1 removes phosphates from target proteins to downregulate several tyrosine kinase-regulated pathways. In hematopoietic cells, the amino-terminal SH2 domain of SHP-1 binds to tyrosine phosphorylated erythropoietin receptors (EpoR) to negatively regulate hematopoietic growth. Overexpression of SHP-1 in epithelial cells results in dephosphorylation of the Ros receptor tyrosine kinase and subsequent downregulation of Ros-dependent cell proliferation and transformation. Following ligand binding in myeloid cells, SHP-1 associates with the IL-3R beta chain and downregulates IL-3-induced tyrosine phosphorylation and cell proliferation.
