Immunofluorescent analysis of SOX2 (green) showing nuclear staining of NCCIT cells (right panel) compared to negative HeLa cell control (left panel). The cells were fixed with formalin for 15 minutes, permeabilized with 0.1% Triton X-100 in TBS, washed, and then blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with a DyLight 488-conjugated SOX2 monoclonal antibody (Product # MA1-014-D488) in 3% BSA-PBS at a dilution of 1:100 and incubated for 1 hour at 37C in the dark. F-Actin (left panel, red) was stained with DyLight 554 Phalloidin (Product # 21834) and nuclei (left panel, blue) were stained with DAPI. Images were taken at 60X magnification.

This intronless gene encodes a member of the SRY-related HMG-box (SOX) family of transcription factors involved in the regulation of embryonic development and in the determination of cell fate. The product of this gene is required for stem-cell maintenance in the central nervous system, and also regulates gene expression in the stomach. Mutations in this gene have been associated with optic nerve hypoplasia and with syndromic microphthalmia, a severe form of structural eye malformation. This gene lies within an intron of another gene called SOX2 overlapping transcript (SOX2OT).
