Imunofluorescent analysis of SOX2 (red) in Human episomal iPSC cells. The cells were fixed with formalin for 15 minutes, permeabilized with 0.1% Triton X-100 in TBS for 10 minutes, and blocked with 3% Blocker BSA (Product # 37525) for 30 minutes at room temperature. Cells were stained with Dylight 650-conjugated SOX2 monoclonal antibody (right, Product # MA1-014-D650) or Dylight 650-conjugated Mouse IgG1 isotype control antibody (left, Product # MA1-191-D650), at a dilution of 1:50 and incubated for 1 hour at room temperature. Nuclei (both panels, blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken at 20X magnification.

This intronless gene encodes a member of the SRY-related HMG-box (SOX) family of transcription factors involved in the regulation of embryonic development and in the determination of cell fate. The product of this gene is required for stem-cell maintenance in the central nervous system, and also regulates gene expression in the stomach. Mutations in this gene have been associated with optic nerve hypoplasia and with syndromic microphthalmia, a severe form of structural eye malformation. This gene lies within an intron of another gene called SOX2 overlapping transcript (SOX2OT).
