Western blot analysis of Sequestosome-1 recombinant protein (lane 1, 200ng) and endogenous Sequestome-1 in extracts from HeLa cells treated with 60 mM chloroquinone for 16 hours (lane 2) using a Sequestome-1 recombinant rabbit oligoclonal antibody (Product # 701510) at a concentration of 1ug/mL. A rabbit anti-tubulin antibody was blotted as a loading control and the blots were developed using chemiluminescence (ECL) method with a goat anti-rabbit HRP-conjugated secondary antibody (Product # G21234). Results show a band at ~62kDa.

ABfinity™ recombinant antibodies are rabbit monoclonal antibodies, unmatched for producing superior results. ABfinity™ antibodies are developed by immunizing animals, screening for functionality, cloning the immunogen-specific antibody genes into high-level mammalian expression vectors, produced on a large scale, and purified with Protein A.
ABfinity™ monoclonal antibodies resemble rabbit monoclonals isolated from serum or produced by hybridomas, but demonstrate greater specificity and sensitivity. Because ABfinity™ recombinant antibodies are derived from cloned DNA sequences of the heavy and light antibody chains, they are not susceptible to cell-line drift or lot-to-lot variation, thus allowing for peak specificity and performance.
Intact IgG appears on a non-reducing gel as ~150 kDa band and upon reduction generating a ~25 kDa light chain band and a ~50 kDa heavy chain.
p62 is a scaffold protein which plays an important role in autophagy by acting a link in recognizing ubiquinated targets and presenting them to autophagic machinery. The N-terminus of p62 aids in polymerization, while the C-terminus aids in binding to ubiquinated proteins and to autophagosomal proteins like LC3. p62 is ubiquitously expressed in adult tissues. It co-localizes with ubiquinated proteins in protein bodies found in the cytosol and nucleus, or within autophagosomes and lysosomal structures.
