Western blot analysis of Sialosyl-Tn antigen was performed by loading 20ug of human tissue lysates from the frontal cortex of 63-year-old (lane 1) and 85-year-old (lane 2) aged brains, per well onto a 10% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat dry milk in PBST for 1 hour at room temperature. The membrane was probed with a Sialosyl-Tn antigen monoclonal antibody (Product # MA1-90577) at a dilution of 1:75 overnight at 4C, washed in PBST, and probed with an HRP-conjugated anti-mouse IgG secondary antibody at a dilution of 1:3500 for 2 hours at room temperature. Sialosyl Tn antigen also known as CD175s is expressed in the majority of colon, gastric, breast, and lung carcinoma. The sialosyl Tn has limited expression to being absent in normal colon, pancreas, stomach, ovary, and prostate. This antigen is detected in several adenocarcinomas. It appears to be useful marker for poorly differentiated colon adenocarcinomsa. This antibody stains colon carcinomas and ovarian mucinous carcinomas producing apical cytoplasmic, apical membrane and luminal staining patterns. It is negative with majority of adult tissue sections.
