Immunofluorescent analysis of Spi2A (green) showing staining in the cytoplasm and nucleus of Jurkat cells (right) compared to a negative control without primary antibody (left). Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with a Spi2A monoclonal antibody (Product # MA1-030) in 3% BSA-PBS at a dilution of 1:20 and incubated overnight at 4°C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight-conjugated secondary antibody in PBS at room temperature in the dark. Nuclei were stained with Hoechst or DAPI (blue). Images were taken at a magnification of 60x.

MA1-030 detects Spi2A in human, mouse and hamster samples.
MA1-030 has been used successfully in Western blot, ICC/IF and ELISA procedures. By Western blot this antibody detects a ~49 kDa band representing the Spi2A protein.
The MA1-030 immunogen is a synthetic peptide corresponding to residues N(418) P E R S M N
F P N G E G A S S Q R L C(436) of hamster Spi2A protein.
Serine protease inhibitor 2A, or Serpin 2A (spi2A), inhibits lysosomal executioner proteases, thus enhancing the survival of cytotoxic T lymphocytes and allowing them to develop into memory CD8 T cells. Through its function of preserving T cells, spi2A has become a research target for such topics as immunity, tumor cell, and AIDS research.
