Immunofluorescent analysis of Synaptojanin 1 (green) showing staining in the cytoplasm of U-87 MG cells (right) compared to a negative control without primary antibody (left). Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with a Synaptojanin 1 monoclonal antibody (Product # MA3-936) in 3% BSA-PBS at a dilution of 1:50 and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight-conjugated secondary antibody in PBS at room temperature in the dark. F-actin (red) was stained with a flourescent red phalloidin and nuclei (blue) were stained with Hoechst or DAPI. Images were taken at a magnification of 60x.

MA3-936 detects Synaptojanin 1 from rat and human samples.
MA3-936 has been successfully used in Western blotting, IHC (frozen), IP and ICC/IF procedures. By Western blot MA3-936 detects a band at 170 kDa representing Synaptojanin 1.
The MA3-936 immunogen is proline rich domain of a fusion protein comprising aa 1156-1286 of rat synaptojanin 1.
Synaptojanin 1 is a phosphatase expressed predominantly in the nervous system. It is concentrated in the nerve terminals where it has been implicated in synaptic vesicle recycling and actin function. There is also evidence that synaptojanin 1 is involved in clathrin-mediated endocytosis.
