Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is pancreas or neuroendocrine tumor.
This antibody recognizes a protein of 38kDa, identified as synaptophysin. It labels normal neuroendocrine cells of human adrenal medulla, carotid body, skin, pituitary gland, thyroid, lung, pancreas, gastrointestinal mucosa, Paneth and quote;s cells in the gastrointestinal tract and of gastric parietal cells. Neurons in the brain, spinal cord, and retina are also labeled. Anti-synaptophysin antibody reacts with neuroendocrine neoplasms of neural as well as epithelial types e.g. neuroblastomas, ganglioneuroblastomas, ganglioneuromas, pheo-chromocytomas, chromaffin, and non-chromaffin paragangliomas.
