Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 10 min at room temperature. A suggested positive control is placenta or kidney tissue.
Fibroblast growth factor-inducible 14 (Fn14) is the receptor for Tweak, a member of the TNF receptor family. It was first described as a weak inducer of apoptosis in some cell types. It is thought to promote angiogenesis and the proliferation of endothelial cells as well as modulating cellular adhesion to matrix proteins. Fn14 is highly expressed in glioma and some cancer cases of breast, pancreas, and liver.
