Ubiquitin is a small regulatory protein predominantly known for its role in protein degradation via the ubiquitin-proteasome pathway. Post-translational modifications of ubiquitin, such as polyubiquitination, involve linking ubiquitin molecules through one of its seven lysine residues (K6, K11, K27, K29, K33, K48, and K63). The K27-linked ubiquitin chains are of particular interest due to their involvement in non-proteolytic functions, including DNA damage response, transcriptional regulation, and immune response. K27-linked polyubiquitination can signal for the recruitment of specific binding proteins that recognize this distinct linkage, influencing various cellular processes. This specific linkage is structurally distinct from other types of ubiquitin chains, forming a compact and rigid conformation that determines its unique binding properties and functional outcomes.
