VAV1 Antibody in Western Blot (WB) — Western blot was performed using Anti-VAV1 Monoclonal Antibody (2E11) (Product # MA5-17198) and a 98kDa band corresponding to VAV1 was observed in Raji, MOLT-4, Jurkat, THP-1 but not in T-47D and MCF7 which are reported to be negative. Whole cell extracts (30ug) of Raji (Lane 1), MOLT-4 (Lane 2), Jurkat (Lane 3), THP-1 (Lane 4), T-47D (Lane 5) and MCF7 (Lane 6) were electrophoresed using NuPAGE® 10 % Bis-Tris gel (Product # NP0302BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).

VAV1 Antibody in Flow Cytometry (Flow) — Flow cytometric analysis of HeLa cells using VAV1 monoclonal antibody (Product # MA5-17198) (green) and negative control (red).

VAV1 Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell lines owing to their inherent genetic constitution. Relative expression of SUFU was observed in Raji, MOLT-4, Jurkat, THP-1, and not in T-47D and MCF7 using VAV1 Monoclonal Antibody (2E11) (Product # MA5-17198) in western blot. {RE}

Target Information

This gene encodes the coiled-coil containing protein optineurin. Optineurin may play a role in normal-tension glaucoma and adult-onset primary open angle glaucoma. Optineurin interacts with adenovirus E3-14.7K protein and may utilize tumor necrosis factor-alpha or Fas-ligand pathways to mediate apoptosis, inflammation or vasoconstriction. Optineurin may also function in cellular morphogenesis and membrane trafficking, vesicle trafficking, and transcription activation through its interactions with the RAB8, huntingtin, and transcription factor IIIA proteins. Alternative splicing results in multiple transcript variants encoding the same protein.
