c-Fos Antibody in Immunocytochemistry (ICC/IF) — Immunocytochemical analysis of c-Fos in HepG2 cells using a c-Fos monoclonal antibody (Product # MA5-36080) at a dilution of 1:27. The cells were treated with 30mM sodium butyrate for 4 hours and counter stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the primary antibody overnight at 4°C. The secondary antibody was an Alexa Fluor 488-congugated Goat Anti-Rabbit IgG(H+L).

c-Fos Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunocytochemical analysis of paraffin-embeded c-Fos in human adrenal gland tissue using a c-Fos monoclonal antibody (Product # MA5-36080) at a dilution of 1:81. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). The section was blocked with 10% normal goat serum for 30 min at room temperature. The primary antibody, in 1% BSA, was incubated at 4°C overnight. A biotinylated secondary antibody was used with an HRP conjugated SP system.

c-Fos Antibody in Western Blot (WB) — Western Blot analysis of Phospho-AKT1 (Thr450) in Hela cells using a Phospho-AKT1 (Thr450) monoclonal antibody (Product # MA5-36079) at a concentration of 2.25 µg/ml. A goat anti rabbit polyclonal secondary antibody was used at a dilution of 1:50,000.

c-Fos Antibody in Flow Cytometry (Flow) — Flow cytometric analysis of c-Fos in Hela cells using a c-Fos monoclonal antibody (Product # MA5-36080) at a dilution of 1:50 is shown in red. The cells were fixed with 70% Ethylalcohol for 18 hours and then permeabilized with 0.3% Triton X-100 for 2 min. The cells were then incubated in 1x PBS /10% normal goat serum to block non-specific protein-protein interactions followed by the primary antibody for 1 h at 4°C. A secondary FITC goat anti-rabbit IgG (H+L) antibody was used at 1/200 dilution for 1 hour at 4°C. The control, indicated in green, was used under the same conditions. An acquisition of >10,000 events was performed.

Target Information

FBJ murine osteosarcoma viral oncogene homolog is commonly known as Cellular protooncogene Fos (c-Fos). It belongs to the fos transcription factor family and is encoded by the C-FOS gene located on chromosome 14. c-Fos heterodimerizes with c-Jun to form the Activator Protein -1 (AP-1) transcription factor which regulates transcription of several genes that play an important role in cellular signal transduction, cell proliferation and differentiation. c-Fos has been shown to be phosphorylated by various stimuli including growth factors and insulin at atleast seven different sites. The best known upstream kinase for phosphorylation at Threonine 232 is ERK MAPK which regulates localization of c-Fos to the nucleus and is thus important for c-Fos induced transcriptional activity.
