Immunofluorescence analysis of c-Kit / CD117 was performed using 70% confluent log phase Ramos cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with c-Kit / CD117 Rabbit Polyclonal Antibody (PA5-16458) at 2µg/ml in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjµgate (A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (S36938). F-actin (Panel c: red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing cytoplasmic localization. Panel e shows the no primary antibody control. The images were captured at 60X magnification.

PA5-16458 targets CD117 in IHC (P) applications and shows reactivity with Human samples.
The PA5-16458 immunogen is a synthetic peptide from C-terminus of human CD117/c-kit protein.
c-kit is a transmembrane receptor tyrosine kinase. It binds to stem cell factor (SCF). It is expressed in many tissues and cells. C-kit/SCf is involved in development of several lineages of stem cells, such as germ cells, neural crest-derived melanocytes and hematopoietic precursor cells Antibody to CD117/c-kit may be helpful in the histopathological identification of gastrointestinal stromal tumors (GIST).

Immunohistochemical clue for the histological overlap of salivary adenoid cystic carcinoma and polymorphous low-grade adenocarcinoma.
PA5-16458 was used in immunohistochemistry to study the potential of immunohistochemistry to distinguish alivary adenoid cystic carcinoma and polymorphous low-grade adenocarcinoma
