Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is breast carcinoma or skeletal muscle tissue.
c-erbB-4 is the fourth member of class 1 receptor kinase family. HER-4 is most predominantly expressed in several breast carcinoma cell lines, and in normal skeletal muscle, heart, pituitary, brain, and cerebellum. Breast tumor cell lines T47-D, MDA-MB-453, BT-474 and H3396 are found to have the highest levels of mRNA, and intermediate levels are seen in MCF-7, MDA-MB-330 and MDA-MB-361. Expression of erbB-4 is low or absent in some breast tumor cell lines such as MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3.
IP-MS enrichment of ERBB4 (Intensity): ERBB4 was enriched 11285-fold from HEK293 lysate compared to background proteins, using the optimized IP-MS workflow with Pierce MS-Compatible Magnetic IP Kit protein A/G (Part No. 90409) and ERBB4 antibody (Part No. PA5-32347).
