Immunohistochemistry analysis of p27Kip1 showing staining in the nucleus of paraffin-embedded human prostate tissue (right) compared to a negative control without primary antibody (left). To expose target proteins, antigen retrieval was performed using 10mM sodium citrate (pH 6.0), microwaved for 8-15 min. Following antigen retrieval, tissues were blocked in 3% H2O2-methanol for 15 min at room temperature, washed with ddH2O and PBS, and then probed with a p27Kip1 Rabbit Polyclonal Antibody (PA516717) diluted in 3% BSA-PBS at a dilution of 1:100 for 1 hour at 37°C in a humidified chamber. Tissues were washed extensively in PBST and detection was performed using an HRP-conjugated secondary antibody followed by colorimetric detection using a DAB kit. Tissues were counterstained with hematoxylin and dehydrated with ethanol and xylene to prep for mounting.

Recognizes a protein of 27kDa, identified as p27Kip1. p27Kip1 is a candidate tumor suppressor gene and functions as a negative regulator of G1 progression and has been proposed to function as a possible mediator of TGF-b induced G1 arrest.
IP-MS enrichment of p27 Kip1 (LFQ intensity): p27 Kip1 was enriched 5386-fold from MCF7 lysate compared to background proteins, using the optimized IP-MS workflow with Pierce MS-Compatible Magnetic IP Kit protein A/G (Part No. 90409) and p27 Kip1 antibody (Part No. PA5-16717).
