LNPs encapsulate mRNAs, protecting them from degradation before the uptake by target cells or tissues. The identification, ratio and purity of the lipid components in LNP formulation are regarded as critical quality attributes (CQAs) for safety and efficacy, and thus must be accurately monitored.<\/p>\n<h3 id=\"toc-hId-2041822799\"><strong>Why do you need a charged aerosol detector?<\/strong><\/h3>\n<p>Traditional UV detection can\u2019t be used, as lipids have no chromophore. A <a href=\"https:\/\/www.thermofisher.com\/document-connect\/document-connect.html?url=https%3A%2F%2Fassets.thermofisher.com%2FTFS-Assets%2FCMD%2Fbrochures%2FBR-70735-LC-Corona-Veo-CAD-BR70735-EN.pdf\" target=\"_blank\" rel=\"nofollow noopener\">charged aerosol detector<\/a> (CAD) generates uniform signal response for any non- or semi-volatile compound, making it an ideal system for LNP compositional analysis. CAD also is more sensitive than alternative lipid detection systems, such as refractive index or evaporative light scattering detectors (ELSD). Over the past few years, charged aerosol detection has been adopted as the standard technology for COVID-19 vaccine release assays at both Moderna and Pfizer\/BioNTech.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignright wp-image-12409\" src=\"http:\/\/admin.acceleratingscience.com\/analyteguru\/wp-content\/uploads\/sites\/25\/2023\/06\/Vaccine-Banner-032322-1.jpg\" alt=\"\" width=\"425\" height=\"91\" srcset=\"https:\/\/admin.acceleratingscience.com\/analyteguru\/wp-content\/uploads\/sites\/25\/2023\/06\/Vaccine-Banner-032322-1.jpg 400w, https:\/\/admin.acceleratingscience.com\/analyteguru\/wp-content\/uploads\/sites\/25\/2023\/06\/Vaccine-Banner-032322-1-300x65.jpg 300w\" sizes=\"auto, (max-width: 425px) 100vw, 425px\" \/>Sissi White, senior product applications specialist at Thermo Fisher Scientific, recently developed a simple UHPLC-CAD method for the characterization and quantification of lipid components of LNP formulations.<\/p>\n<p>With the method:<\/p>\n<ul>\n<li>Baseline separation was achieved for lipids and their impurities within 10 minutes, using a Thermo Scientific\u2122 <a href=\"https:\/\/www.thermofisher.com\/order\/catalog\/product\/27826-252130?SID=srch-srp-27826-252130\" target=\"_blank\" rel=\"nofollow noopener\">Accucore\u2122 C30 column<\/a> on a <a href=\"https:\/\/www.thermofisher.com\/order\/catalog\/product\/IQLAAAGABHFAPUMBHV?SID=srch-hj-IQLAAAGABHFAPUMBHV\" target=\"_blank\" rel=\"nofollow noopener\">Thermo Scientific\u2122 Vanquish\u2122 Flex UHPLC system<\/a>.<\/li>\n<li>Sensitive and reproducible detection was obtained for all lipids, including cholesterol, with detection limits at ~10 ug\/mL, using a <a href=\"https:\/\/www.thermofisher.com\/order\/catalog\/product\/VF-D20-A?SID=srch-hj-VF-D20-A\" target=\"_blank\" rel=\"nofollow noopener\">Vanquish charged aerosol detector<\/a>.<\/li>\n<\/ul>\n<p><span class=\"lia-inline-image-display-wrapper lia-image-align-center\" style=\"width: 999px\"><span class=\"lia-inline-image-caption\"><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-12413 aligncenter\" src=\"http:\/\/admin.acceleratingscience.com\/analyteguru\/wp-content\/uploads\/sites\/25\/2023\/06\/Chromatograms-using-FA-method-032322-1.png\" alt=\"\" width=\"998\" height=\"464\" srcset=\"https:\/\/admin.acceleratingscience.com\/analyteguru\/wp-content\/uploads\/sites\/25\/2023\/06\/Chromatograms-using-FA-method-032322-1.png 998w, https:\/\/admin.acceleratingscience.com\/analyteguru\/wp-content\/uploads\/sites\/25\/2023\/06\/Chromatograms-using-FA-method-032322-1-300x139.png 300w, https:\/\/admin.acceleratingscience.com\/analyteguru\/wp-content\/uploads\/sites\/25\/2023\/06\/Chromatograms-using-FA-method-032322-1-768x357.png 768w\" sizes=\"auto, (max-width: 998px) 100vw, 998px\" \/>Figure 3.
