Repairing damaged tissues, treating once-intractable diseases; cell therapy poses profound possibilities for situations once thought to be beyond the reach of contemporary science.<\/p>\n<p>By 2001, scientists had employed cell therapy technology to not only create the first beating heart cells outside the human body,<sup>1<\/sup> but also to bestow upon the world a zoological marvel: Dolly, the world\u2019s most famous sheep.<sup>2 <\/sup>The burgeoning field\u2019s capacity to shift the landscape of health technologies seems boundless.<\/p>\n<p>However, cell therapy isn\u2019t without its practical challenges. By replacing lost or damaged material,<\/span><span class=\"NormalTextRun SCXW219062476 BCX0\"> viable cells <\/span><span class=\"NormalTextRun SCXW219062476 BCX0\">may present therapeutic relief in the treatment of a disease, neurological disorder, or major accident.\u00a0<\/span><\/span><span class=\"EOP SCXW219062476 BCX0\" data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<h2><span class=\"TextRun SCXW52179895 BCX0\" lang=\"EN-US\" xml:lang=\"EN-US\" data-contrast=\"auto\"><span class=\"NormalTextRun SCXW52179895 BCX0\">How does it work?\u00a0<\/span><\/span><span class=\"EOP SCXW52179895 BCX0\" data-ccp-props=\"{}\">\u00a0<\/span><\/h2>\n<p><span data-contrast=\"auto\">In the 19<\/span><span data-contrast=\"auto\">th<\/span><span data-contrast=\"auto\"> century, physiologist Charles-\u00c9douard Brown-S\u00e9quard<sup>3<\/sup> sought to reverse the symptoms of aging with injections of animal cells.<\/span><span data-contrast=\"auto\">iii<\/span><span data-contrast=\"auto\"> Modern cell therapy has developed from a similar principle \u2013\u202ftreatment from the introduction of intentionally cultivated cells \u2013 but with a more precise scope.\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<h3><span data-contrast=\"auto\">Allotransplantation<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/h3>\n<p><span data-contrast=\"auto\">If you\u2019ve ever donated blood, you may have taken part in allotransplantation. In this method, genetic material \u2013 cells, muscles, organs \u2013\u202fis taken from one organism and transplanted into a non-genetically identical member of the same species. For humans, this is often seen in blood transfusions and organ implants.\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">The transplanted material, known as an allograft or allogeneic transplant, may trigger immunosuppressive responses from the body. For this reason, commercially available products such as <\/span><a href=\"https:\/\/www.thermofisher.com\/us\/en\/home\/clinical\/cell-gene-therapy\/cell-gene-therapy-learning-center\/cell-gene-therapy-resource-library\/cell-therapy-handbook\/overview-cell-isolation-engineering-expansion.html?icid=bid_cbu_sbu_r01_co_CP1450_PJT9002_bid11111_0so_blg_op_awa_kt_s00_celltherapy1\"><span data-contrast=\"none\">allogeneic CAR T cell therapies<\/span><\/a><span data-contrast=\"auto\">, require conscientious gene editing technique to minimize the risks of immunosuppressive rejection.\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">Allogeneic CAR T cell therapies utilize a chimeric antigen receptor (CAR) engineered to be expressed on immune cells such as T and NK cells. This allows for the targeting of specific cancerous cells.\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">Allogeneic CAR T cells, as a singular product that can be used for multiple patients, offer a wider range of application than autologous CAR T cells. By editing multiple genes, allogeneic CAR T cell products can be configured for multiple cancer targets.\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-ccp-props=\"{}\">\u00a0<\/span><span data-contrast=\"auto\">These products offer a treatment that combines the accessibility of a mass-produced, off-the-shelf product with the focused implantation needed to treat individuals.\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<h3><span data-contrast=\"auto\">Autotransplantation<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/h3>\n<p><span data-contrast=\"auto\">This method is a little more personal. In autotransplantation, cells from one part of an individual organism are transplanted to a different part of that same organism. A very common example of this process happens in hospitals everyday when healthcare workers collect blood from someone expecting a surgery or other major medical intervention.\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">The blood, if correctly stored, is easily re-introduced back to the individual, aiding in their recuperation with an influx of blood that poses minimal risk of rejection.<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">The ease with which <\/span><a href=\"https:\/\/www.thermofisher.com\/us\/en\/home\/global\/forms\/car-t-cell-manufacturing-white-paper.html?icid=bid_cbu_sbu_r01_co_CP1450_PJT9002_bid11111_0so_blg_op_awa_kt_s00_celltherapy7\"><span data-contrast=\"none\">autologous CAR T cell therapies<\/span><\/a><span data-contrast=\"auto\"> bypass potentially fatal immunosuppressive responses present a clear advantage for this method in cases when applicable.\u00a0 However, the conditions that afford autologous that advantage also pose technical limitations to its scalability.\u00a0<\/span><\/p>\n<h2><span data-contrast=\"auto\">What Would Cells in Therapy Share if they Could?<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/h2>\n<p><span data-ccp-props=\"{}\">\u00a0<\/span><span data-contrast=\"auto\">Cell therapies are beset by several practical considerations that pose limits on more global applications of the technology.\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">The intended outcomes of cell therapy \u2013 whether it\u2019s treating cancer or rehabilitating an athletic injury \u2013\u202faren\u2019t accomplished with one or two or even ten good cells. It takes many, many cells, transplanted over the course of potentially many treatments, for long-term therapeutic benefit.\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">This can be especially problematic in situations where autologous cells are required: an organism only produces so many cells over the course of their lifetime. Viable cells intended for treatment must be produced at a rate exponentially significant enough to fulfill the intended treatment.\u00a0<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<p><span data-ccp-props=\"{}\">\u00a0<\/span><span data-contrast=\"auto\">Cultivating viable cells for transplant is time-consuming.
