You might also struggle to get the image quality you&rsquo;re expecting. Yet with the right sample preparation techniques, you can easily improve your results.<\/p>\n<h2 style=\"font-size:100%\"><strong>High moisture samples<\/strong><\/h2>\n<p>First things first: SEMs operate in&nbsp;a&nbsp;vacuum,which can seriously alter your sample.&nbsp;For example, loose particles can&nbsp;detach from any surface, all liquids&nbsp;will&nbsp;evaporate immediately,&nbsp;and delicate materials will outgas.<\/p>\n<div style=\"width: 410px\" class=\"wp-caption alignmiddle\"><img loading=\"lazy\" decoding=\"async\" src=\"https:\/\/admin.acceleratingscience.com\/materials\/wp-content\/uploads\/sites\/7\/2022\/04\/sample20prep1.jpg\" style=\"vertical-align: middle;margin: 10px\" alt=\"SEM image of a fly eye requiring specialized sample preparation\" title=\"SEM image of a fly eye requiring specialized sample preparation\" width=\"400\" height=\"423\" \/><p class=\"wp-caption-text\">SEM image of a fly's eye<\/p><\/div>\n<p><span>While a fly&nbsp;can be easily imaged, when you image<\/span><span> a&nbsp;fly larva, you will most likely get&nbsp;a splatter&nbsp;scenario.&nbsp;That&rsquo;s because larvae can be understood as small liquid pockets. If the water inside evaporates, the skin can collapse due to the internal pressure and, in the worst cases, it might explode.&nbsp;Gasses&nbsp;and particles&nbsp;can&nbsp;then access the electron column and compromise image quality for good.<\/span><\/p>\n<p><span>To image high-moisture content samples with an SEM, it&rsquo;s critical to:<\/span><\/p>\n<ul>\n<li><span>Operate at a lower vacuum level<\/span><\/li>\n<li><span>Freeze your sample<\/span><\/li>\n<li><span>Dry your sample<\/span><\/li>\n<\/ul>\n<p><span><div style=\"width: 410px\" class=\"wp-caption alignmiddle\"><img loading=\"lazy\" decoding=\"async\" src=\"https:\/\/admin.acceleratingscience.com\/materials\/wp-content\/uploads\/sites\/7\/2022\/04\/sample20prep2.jpg\" style=\"vertical-align: middle;margin: 10px\" alt=\"SEM image of an apple\" title=\"SEM image of an apple\" width=\"400\" height=\"425\" \/><p class=\"wp-caption-text\">Fig. 2: SEM image of an apple at -25\u00b0C<\/p><\/div><\/span><\/p>\n<h2 style=\"font-size:100%\"><strong>Non-conductive samples in SEM<\/strong><\/h2>\n<p><span>In addition to the vacuum, you should also consider the effect of electrons accumulating on your sample. This translates into an image with bright areas where all the details can no longer be observed.
