The ProtoArray® Human Protein Microarray is an advanced, high-content, functional protein microarray that enables rapid profiling of thousands of biochemical interactions in as little as one day. Researchers can use the ProtoArray® microarray to perform a variety of assays including biomarker identification, drug target discovery, enzyme substrate identification, antibody specificity profiling, and protein-protein interaction studies. Results can be readily measured with most commercial scanners, and the data can be analyzed using our free ProtoArray® Prospector software (Figure 1).

Absolute identification of your protein targets with ProtoArray® Human Protein Microarray

Each ProtoArray® protein array has over 9,000 human proteins printed in duplicate on a one inch by three inch slide. The proteins are produced from clones selected from the Invitrogen Ultimate™ ORF Collection, the first and only clone resource to offer full-insert sequenced clones in a Gateway® vector. Proteins are expressed as N-terminal GST fusion proteins using a baculovirus expression system. After protein expression, proteins are purified under nondenaturing conditions. The proteins are printed at +4°C on an ultrathin layer of nitrocellulose to preserve native protein structure without the need for chemical fixation. Each slide is labeled with a unique bar code for easy tracking.

Individual microarrays on nitrocellulose-coated glass slides allow for the greatest flexibility in experimental design, with protocols for each supported application available. Some of these profiling applications (Figure 2) include:

Profiling with ProtoArray® Protein Microarrays

ProtoArray® Human Protein Microarrays enable a broad range of research applications and are available both as complete optimized kits for certain applications as well as individual arrays to support a broader range of applications. Complete kits with optimized product configurations for screening protein-protein interactions and kinases against thousands of potential native protein substrates are available. Protein-protein interaction studies can be completed in as little as one day using either a V5-tagged or biotinylated protein probe with Anti-V5-Alexa Fluor® 647 Antibody or streptavidin-conjugated Alexa Fluor® 647 for detection. Screening of kinases can be completed in as little as 2 days with no need for special instrumentation—detection can be done with either a phosphorimager or conventional autoradiography film.

Using the ProtoArray® Human Protein Microarray is simple, and you get high-quality results in a fraction of the time. Regardless of your study, fluorescent applications can be completed in as little as one day. ProtoArray® microarray applications follow the same simple workflow of incubating a sample on the array, probing with a detection reagent, and then visualizing with a fluorescent scanner or phosphorimager (Figure 3).
