Essential 8 Flex Medium provides all of the benefits of Essential 8 Medium; reduced variability of components, reliable and robust cultures with a xeno-free, cGMP produced media; along with the added benefit of flexibility. With Essential 8 Flex Medium, your pluripotent stem cell maintenance schedule is no longer controlled by the need to be in the lab every day to change the culture medium.
With Essential 8 Flex Medium, PSCs need to be fed only on the day of splitting and the day after, if the cells are to be left without feeding for more than 48 hours (the weekend), double the feed volume on Friday.
This workflow has been carried out for over 30 passages with cultures maintaining pluripotency, trilineage potential, and normal karyotype. Essential 8 Flex Medium, based upon the published formulation of Essential 8 Medium by Chen et al., allows for weekend-free pluripotent stem cell culture without a negative impact to the health of the stem cells.

CTS Immune Cell Serum Replacement (SR) for T cell culture
Induced pluripotent stem cells (iPSCs) can be differentiated into many different cell types including human T lymphocytes where they have shown promise in providing an unlimited source of antigen-specific T cells targeting cancer.1These iPSC-derived T cells and T cells isolated from peripheral blood mononuclear cells (PBMCs) are expanded in vitro and typically require human serum–supplemented media, which can lead to variability and supply concerns. We have developed Gibco CTS Immune Cell SR, which can replace human serum in immune cell culture including T cell culture. CTS Immune Cell SR has been shown to support equivalent expansion of functional CART-19 T cells compared to human serum–supplemented media as demonstrated in in vitro expansion assays. A leukemia model in immunodeficient mice was used to determine the efficacy of CD19-specific CAR T cells grown in medium supplemented with CTS Immune Cell SR. CAR T cells were able to control leukemia cells two weeks after infusion, while control T cell–treated mice succumbed to leukemia. CTS Immune Cell SR may be a suitable replacement for human serum in culture media when expanding immune cells including T cells isolated from PBMCs or iPSC-derived T cells.
