NovaBright™ SEAP Enzyme Reporter Gene Assay
Detection of secreted alkaline phosphatase (SEAP) reporter gene activity is easier than ever with the NovaBright™ SEAP 2.0 reporter gene assay. By utilizing our high-performance alkaline phosphatase substrate, CSPD, and our next-generation Emerald™ enhancer, better assay performance is obtained over 5 orders of magnitude of enzyme concentration. The assay has been simplified down to two ready-to-use assay reagents, eliminating reagent preparation and sample dilution steps. Our proprietary buffer formulation ensures low background from endogenous phosphatases, so NovaBright™ SEAP 2.0 enables the most sensitive detection by providing a higher signal-to-noise ratio of SEAP reporter gene activity than other assays of its kind.

BrdU Mouse Monoclonal Antibody, Clone MoBU-1
Cell proliferation can be measured by following the incorporation of the thymidine analog BrdU into newly synthesized DNA, and subsequent detection with an anti-BrdU antibody. The MoBU-1 clone has high specificity for BrdU while displaying no cross-reactivity with EdU, thus making it valuable for use in dual pulse applications. The MoBU-1 clone is available as a direct conjugate for flow cytometry for use with the violet, blue, and red lasers. Unconjugated, biotinylated, and direct conjugates with photostable Alexa Fluor dyes have also been validated for use in immunofluorescence microscopy and high-content applications.
Labeling of cell proliferation using BrdU and BrdU Mouse Monoclonal Antibody conjugates. Jurkat T cell leukemia cells were treated with 10 μM BrdU for 1.5 hr. Cells were then fixed in ethanol and stored overnight at ≤–20°C. An acid denaturation method was used before labeling 1 x 106 cells with 5 μL of the following BrdU Mouse Monoclonal Antibody (Clone MoBU-1) conjugates: (A) Pacific Blue™ conjugate, (B) Alexa Fluor 488 conjugate, and (C) Alexa Fluor 647 conjugate. Proliferating cells are clearly distinguished from nonproliferating cells.
The Premo™ Autophagy Sensor combines the selectivity of an LC3B–fluorescent protein (LC3B-FP) chimera with the transduction efficiency of BacMam technology, enabling unambiguous visualization of this protein. Unlike expression vectors, BacMam reagents enable titratable and reproducible expression and offer high cotransduction efficiency. Recent improvements to the BacMam system enable efficient transduction in a wider variety of cells, including neurons and neural stem cells, with an easy one-step protocol. Each Premo™ Autophagy Sensor kit includes a BacMam LC3B-FP, a control BacMam LC3B (G120A)-FP, and chloroquine diphosphate to artificially induce autophagosome accumulation. The LC3B Antibody Kit for Autophagy includes a rabbit polyclonal antibody against LC3B that has been validated for use in fluorescence microscopy and high-content imaging and analysis. Chloroquine diphosphate is also included; following treatment with this compound, normal autophagic flux is disrupted and autophagosomes accumulate as a result of increased lysosomal pH.

The Frizzled family of transmembrane proteins serves as receptors for the Wnt signaling pathway. Wnt signaling is involved in various developmental processes, including cell polarity, cell fate determination, and the control of cell proliferation.
