By combining improved source/optics, higher detector performance, and reduced contamination/drift, the work argues that 100 keV cryo-TEMs can be viable for routine, high-resolution structure determination, potentially lowering cost/complexity barriers for labs adopting cryo-EM.
Researchers report an open, community resource for visual proteomics: 1,829 annotated in situ cryo-electron tomography (cryo-ET) tomograms plus raw data from the green alga Chlamydomonas reinhardtii. Using cryo-plasma FIB milling and modern cryo-ET workflows, they capture detailed views of diverse eukaryotic organelles and benchmark dataset quality by subtomogram averaging of native complexes (including ribosomes, Rubisco, nucleosomes, microtubules, clathrin, photosystem II, and ATP synthase), with many reconstructions reaching sub-nanometer resolution. The dataset is designed to accelerate both method development (segmentation, averaging, analysis) and biological discovery by making large-scale, high-quality cryo-ET data broadly reusable.
