Growth of frozen Chlamydomonas reinhardtii

Three Chlamydomonas reinhardtii cultures were grown at 50 µE m-2s-1, 26°C, and 110 rpm, and optical densities were measured at OD750 at 16, 24, 40, 48, 64, and 80 hrs.  The culture represented by the blue line started with a fresh culture, and the cultures represented by the red and green lines were frozen at different cell concentrations using the GeneArt® Cryopreservation Kit for Algae and resuscitated in TAP growth media, following manufacturer recommendations.   Similar results were seen with Chlorella vulgaris, either wild type or transformants.
