The Human RANTES(Hu regulated upon activation, normal T-cell expressed and secreted)) ELISA quantitates Hu RANTES in human serum, plasma, cell culture supernatants, amniotic fluid or other body fluids. The assay will exclusively recognize both natural and recombinant Hu RANTES.
Principle of the method
The Human RANTES solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the binding of the second (detector) antibody to the target on a different epitope from the capture antibody. An antibody conjugated with enzyme binds the formed sandwich. After incubation and washing steps to rid the microplate of unbound substances, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.
Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. See manual for more information on validation.
RANTES is a chemoattractant for blood monocytes, memory T helper cells and eosinophils. It causes the release of histamine from basophils and activates eosinophils. It binds to CCR1, CCR3, CCR4 and CCR5 and is one of the major HIV suppressive factors produced by CD8+ T cells. Recombinant RANTES protein induces a dose dependent inhibition of different strains of HIV 1, HIV 2, and simian immunodeficiency virus (SIV). It is T cell and macrophage specific, induced by mitogens, and belongs to the intercrine beta (chemokine CC) family.
