The Human TACE/ADAM17 ELISA quantitates Hu TACE in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu TACE.
Principle of the method
The Human TACE solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

ADAM17 (disintegrin and metalloproteinase domain-containing protein 17) cleaves the membrane bound precusor of TNF-alpha to its mature soluble form. ADAM 17 is responsbile for the proteolytical release of soluble JAM3 from endothelial cells surface and the proteolytic release of several other cell-surface proteins, including p75 TNF-receptor, interleukin receptor type II, p55 TNF-receptor, transforming growth factor-alpha, L-selectin, growth hormone receptor, MUC1, and the amyloid precursor protein. It acts as an activator of the Notch pathway by mediating cleavage of Notch and generating the membrane-associated intermediate fragment called Notch extracellular truncation (NEXT). Mutations in the gene can result in inflammatory skin and bowel disease, neonatal 1.
