Topic: Understanding the genetic basis of tumor formation and heterogeneity in FFPE and frozen cancer samples using LCM and PGM
Abstract: Identifying mutations and transcript abundance in tumor for understanding its molecular mechanism and ultimately the design of treatments are often used. Microtome equipped with Section Transfer System (STS) and Cryostar NX70 Automated Cryostat minimizes damage to tissue and produces thin and high quality sections. Analysis of whole tumor sections may provide misleading results, and Laser Capture Microdissection (LCM) is the only mechanism that can isolate single cells from slides while preserving surrounding cells and tissue. Arcturus LCM system offers the powerful combination of infrared capture laser and ultraviolet laser, and allows for capturing either single cells or dense tissue structures. LCM facilitates the efficient analysis by enriching cancer cells. Clinical cancer research specimens are often degraded and difficult to work with. In order to have targeted approach to mutation detection and gene expression analysis, multiplex high-throughput targeted sequencing with Ion PGM system is presented. Ion Ready-to-Use cancer panels using multiplex PCR amplify targets from 10ng DNA from FFPE material. With Oncomine knowledgebase, pathogenic mutations may link to actionable drugs and provide information for studies in a clinical environment.
