Expression and activity data for bacteriorhodopsin using the MembraneMax™ kits

We used reagents from the MembraneMax™ kits to express, purify, and determine the activity of Halobacterium salinarum bacteriorhodopsin (bR), a 7-transmembrane a-helix light-driven proton pump. Retinal substrate binds only to bacteriorhodopsin when it is correctly folded and thus serves as an ideal assay to determine protein activity. With the addition of MembraneMax™ reagent, the color changed from clear (not active) to pink (active) (panel A), and significantly higher solubility was observed (panel B). Additional data not shown here is that bR expresses at 0.9 mg/mL.
