Deeper Human Proteome Profiling with Higher Throughput via Optimized Ion Processing

This webcast introduces the new ultra-sensitive and fast scanning Orbitrap Astral Zoom mass spectrometer for human proteome analysis and single-cell proteomics with unprecedented speed and coverage. This is achieved by taking advantage of the new instrument features including improved MS2 peak deconvolution, increased scan speed, and enhanced ion accumulation for MS2 scans, collectively providing improved performance in proteomics research without sacrificing sensitivity or quantitative integrity.

 

These improvements allow the Orbitrap Astral Zoom MS to exceed acquisition speeds of 270 Hz, with pre-accumulation increasing signal intensity by close to 50%. This enables the identification of > 36,000 unique peptides-per-minute with a 300 samples-per-day (SPD) LC gradient resulting in identification of >113,000 peptide precursors and >8,300 protein-coding genes from a 250ng HEK293 digest. Similar gains are observed for single-cell proteomics for which the optimized methodology and a low-input application identified a median of ~6,600 protein-coding genes from single HeLa cells using a 60-SPD LC method.

Prof. Jesper Olsen studied analytical chemistry and obtained his PhD in biochemistry and molecular biology at University of Southern Denmark. He was involved in developing high-resolution MS-based proteomics. In 2012, he was promoted to vice director of Novo Nordisk Foundation for Protein Research at University of Copenhagen, and in 2014 full professor.
