High Mobility Group Box 1 (HMGB1) recombinant human protein is supplied as a lyophilized powder. This protein is suitable for use in protein studies such as protein structure analysis and protein-protein interactions. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.

This recombinant protein is expressed from a DNA sequence encoding the human HMGB1 protein (NP_002119.1) (Met 1-Glu 215) fused to the fused Fc region of human IgG1 at the N-terminus.

N-terminal Sequence Analysis: Glu 20.

Activity: measured by its binding ability in a functional ELISA. Immobilized recombinant mouse AGER at 2 µg/mL (100 µL/well) has been shown to bind human HMGB1. The EC50 of human HMGB1 is 0.23 µg/mL.

Formulation:

Reconstitution: dissolve the protein in sterile double-distilled water to a concentration of 25 mg/mL or lower. It is recommended that the protein be aliquoted and used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Background
Human High-mobility group box 1 protein (HMGB1), also known as HMG-1 or amphoterin previously, is a member of the HMGB family consisting of three members, HMGB1, HMGB2 and HMGB3. HMGB1 is a non-histone architectural chromosomal protein ubiquitously present in all vertebrate nuclei and binds double-stranded DNA without sequence specificity. HMGB1 is structurally composed of two homologous DNA-binding domains entitled box A and box B and a highly negative-charged C terminus. This protein is extremely conserved across species with 99- 100% aa sequence homology to mouse, rat, bovine, canine, porcine proteins. As a nuclear protein, HMGB1 stabilizes nucleosomes and allows bending of DNA that facilitates gene transcription which is essential for individual survival. Meanwhile, it is revealed that HMGB1 can also act as a cytokine extracellularlly and regulates monocyte, T cell, dendritic cell activities in inflammatory responses. Once secreted, HMGB1 induces signal transduction through its receptor RAGE and perhaps TLR2 and TLR4. The immune-regulatory function of HMGB1 may lead to being identified as new therapeutic target.
