Selectin E (SELE) recombinant human protein is supplied as a lyophilized powder. This protein is suitable for use in analysis of protein structure and performing cell based assays. It can also be used as an immunogen, as a protein standard, or in other research applications.

This recombinant protein is expressed from a DNA sequence encoding the extracellular domain (Met 1-Pro 556) of the human SELE precursor(NP_000441.2) fused to the polyhistidine-tagged Fc region of human IgG1 at the C-terminus.

N-terminal Sequence Analysis: Trp 22.

Activity: measured by the ability of the immobilized protein to support the adhesion of U937 human histiocytic lymphoma cells . When cells are added to SELE coated plates (2 µg/mL, 100 µL/well) approximately 90 %-100 % will adhere specifically .

Formulation: lyophilized in 140 mM NaCl, 2.7 mM KCl, 10 mM Na2HPO4, 1.8 mM KH2PO4, pH 7.4, 5% mannitol, 5% trehalose, 0.02% Tween®-80.

Reconstitution: Dissolve the protein in sterile double-distilled water to a concentration of 0.2 mg/mL or lower. It is recommended that the protein be aliquoted and used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Expiration Date: expires one year from date of receipt when stored as instructed.

Background
E-selectin, also known as endothelial leukocyte adhesion molecule-1 (ELAM-1) and CD62E, is an inducible adhesion molecule that is expressed at relatively high density on the surface of cultured endothelial cells that have been activated by proinflammatory cytokines, such as IL- 1β, TNF-α, or bacterial endotoxin. E-selectin exhibits a complex mosaic structure consisting of a large extracellular region comprised of a lectin domain, an EGF-like domain, and a short consensus repeat (SCR) domain, followed by a transmembrane region and a relatively short (32 aa) cytoplasmic tail. As a member of the LEC-CAM or selectin family, Eselectin recognises and binds to sialylated carbohydrates including members of the Lewis X and Lewis A families found on monocytes, granulocytes, and T-lymphocytes. E-selectin supports rolling and stable arrest of leukocytes on activated vascular endothelium, and furthermore, it was indicated that it can also transduce an activating stimulus via the MAPK cascade into the endothelial cell during leukocyte adhesion. In addition, this adhesion molecule appears to be involved in the pathogenesis of atherosclerosis.
