Cluster of Differentiation 4 (CD4) recombinant human protein is supplied as a lyophilized powder. This protein is suitable for use in the study of protein-protein interactions, and cell biology research applications. It can also be used as a protein standard.

This recombinant human protein is expressed from a DNA sequence encoding the extracellular domain (Met 1-Trp 390) of human CD4 (NP_000607.1) fused to the polyhistidine-tagged Fc region of human IgG1 at the C-terminus.

N-terminal Sequence Analysis: Lys26.

Activity: measured by its binding ability in a functional ELISA. Immobilized human CD4-His at 2 µg/well has been shown to bind biotinylated HIV-1 GP120 with a linear range of 0.078-1.25 µg/mL.

Formulation: lyophilized in PBS, pH 7.4, 5% mannitol, 5% trehalose, and 0.02% Tween®-80.

Reconstitution: Dissolve the protein in sterile double-distilled water to a concentration of 0.2 mg/mL or lower. It is recommended that the protein be aliquoted and used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

Expiration Date: expires one year from date of receipt when stored as instructed.

Human T-cell surface glycoprotein CD4, also known as T-cell surface antigen T4/Leu-3, is a single-pass type I membrane protein. CD4 contains three Ig-like C2-type (immunoglobulin-like) domains and one Ig-like Vtype (immunoglobulin-like) domain. CD4 is a glycoprotein expressed on the surface of T helper cells, regulatory T cells, monocytes, macrophages, and dendritic cells. CD4 amplifies the signal generated by the TCR. CD4 interacts directly with MHC class II molecules on the surface of the antigen presenting cell via its extracellular domain. CD4 is a primary receptor used by HIV-1 to gain entry into host T cells. HIV infection leads to a progressive reduction of the number of T cells possessing CD4 receptors. Therefore, medical professionals refer to the CD4 count to decide when to begin treatment for HIV-infected patients.
