Thermo Scientific Pierce TFCS forms amide bonds with primary amines (e.g., lysine side chains) to add a seven-atom spacer arm (7.7 angstroms) terminated by a trifluoroacetyl group that can be removed later to expose a new primary amine.

TFCS ( N-[ε-trifluoroacetylcaproyloxy]succinimide ester) is an amine-reactive modification reagent with a protected primary amine group. This reagent is used to temporarily block amine groups in target molecules and to extend lysine side chains to reduce steric hindrance. The coupling reaction is performed in phosphate-buffered saline at pH 7.2, which also preserves the amine protecting group (i.e., trifluoroacetyl). The trifluoroacetyl is removed at pH 7.8-8.1 in phosphate or borate buffer.
