Thermo Scientific Precise Tris-HEPES Protein Gels, 12%, 15-Well are precast polyacrylamide gels for protein electrophoresis (PAGE) that use Tris-HEPES running buffer, have long shelf lives and provide compatibility with different commercial gel tanks.

Precise Gels are cast in a durable plastic cassette using a neutral-pH buffer that prevents polyacrylamide breakdown and results in a long shelf life. High-resolution staining and transfer of proteins is accomplished quickly on these 1mm thick mini gels. Gels are individually packaged in an easy-to-open plastic pouch and are ready to run with no comb or tape to remove. Following electrophoresis, the cassettes are easy to dismantle (no special tools required) without damaging the gel.

SDS-PAGE (sodium dodecyl sulfate polyacrylamide gel electrophoresis) is used to determine the approximate molecular weights of proteins with reference to the mobility of standard proteins on the same gel. Gradient gels provide the appropriate gel pore size, making molecular weight estimation more accurate by sharpening stained protein bands.

The most popular buffer system for SDS-PAGE is the Laemmli system [Nature 227, 680-686 (1970)]. This system, however, is limited by extended running times and gel instability from hydrolysis of polyacrylamide to acrylic acid in alkaline conditions. This chemical change alters gel conductivity, causing a constant change in protein migration patterns with time.

Polyacrylamide hydrolysis does not occur at neutral pH, and Thermo Scientific Precise Protein Gels are cast at pH 7, yielding a long shelf life and assured reproducibility of protein migration patterns. The extra stability of Precise Protein Gels is combined with a new Tris-HEPES running buffer to give speed and excellent resolution with the same size ranges as the Laemmli system. The buffer can be made according to the recipe provided in the instruction booklet or purchased premixed.
