What pore size, loading, and type of resin is recommended for syntheses on the 433A?

The Model 433A Peptide Synthesis System is a batch synthesizer. With Fmoc chemistry, the optimal conditions are: 200-400 "mesh" (or 70-40 micrometer diameter) beads are preferred, but 100-200 mesh (larger beads) will work. This assumes the primary solvent is "NMP" (N-methylpyrrolidone). For the standard chemistries, the loading should be about 0.5 to 0.9 mmole/g (Fmoc protected active group/ per g of resin). Use of lower loaded resins may require both a larger reaction vessel and a different chemistry, or may not be advised. The best resin type is HMP (p-hydroxymethylphenoxymethyl) polystyrene resin, which may also be called "Wang" resin. If PEG (polyethylene glycol-polystyrene) or other "extended" resins are used, adjustment must be made to accommodate the typically lower loading, and less favorable swelling properties.
